Nephelometry and photometry
An analytical chemistry technique called nephelometry is used to quantify how cloudy or turbid a solution is as a result of suspended insoluble particles. Light is transmitted, absorbed (blocked), and dispersed as it passes through a turbid solution with suspended solid particles. The size, shape, and concentration of the insoluble particles in solution as well as the wavelength of the input light all affect how much light is scattered. In the nephelometry technique the amount of this scattered light is measured with the help of a nephelometry detector placed at a specific angle (usually at 90 degree). Nephelometry was originally used in immunoassays for the detection and measurement of serum proteins in the blood, such as immunoglobulins and macromolecules, in the field of clinical chemistry.
The amount of light that passes through the sample is measured by photometry. In photometry, a sample containing insoluble particles in solution is illuminated with light of a specified wavelength, and the light that passes through the sample is collected by a detector positioned in line with the light source.
The concentration of the sample and the size of the scattering particles are the two key determinants of whether to use nephelometry or photometry. The intensity of the transmitted light will be about equivalent to the intensity of the light source if the sample includes few insoluble particles (low concentrations). Nephelometry is a better option in this situation. Since light transmission is greatly reduced in samples with a high concentration of insoluble particles, photometry is a better fit.
Difference between conventional nephelometer and new generation nephelometer
Conventional nephelometers works based on endpoint nephelometry and new generation nephelometers work based on rate-based or kinetic nephelometry.
Endpoint (or fixed-time) nephelometry detects the maximum scattered light after an antigen-antibody interaction has achieved equilibrium, or after a defined reaction time. Rate or kinetic nephelometry is an alternate approach that uses repeated observations over time used over the whole precipitation process and often offers more information on the reaction than endpoint nephelometry method.

For all protein tests in blood testing, the Mispa-i2 is a semi-automated specialized protein analyzer that provides excellent accuracy and rapid turnaround times. Mispa i2 helps the clinical treatment of a range of disorders like Diabetes, Cardiovascular risk, Inflammation, etc. by providing a broad test panel of 25 parameters.
The product Mispa i2 is entirely produced in India and aids in quantitative specific protein quantification by combining the industry-recognized nephelometry and turbidimetry approaches. The Mispa i2 uses a special, patented technology to deliver the highest sensitivity and linearity, ensuring high-quality findings.
A decade ago, nearly 35 percent of specific protein estimations were done by the latex method. The latex methods are qualitative/semi-quantitative tests. Due to this limiting factor of latex testing, there was always an uncertainty of results in the borderline areas of reference ranges. These tests were affordable to the rural population but due to its low specificity and sensitivity, the purpose of the testing was eluded in many cases.
Approximately 45 percent of the protein estimations were performed in turbidimetry assays in fully automated or semi-automated systems at a higher cost. Nephelometry, the best method for protein testing, was unreachable to the common laboratories because of the very high cost of the automated instruments, reagents, and consumables.
With the motive to provide high-quality protein estimation at an affordable cost, Agappe introduced this flagship product for protein estimation in 2012. Within a decade, Mispa i2 emerged as the game changer in specific protein estimation.
How does Mispa -i2 differ from other protein analyzers?
The dual measuring technology (rate-based nephelometry and immunoturbidimetry) with the unique patented technology - UCS (Unique Channel Shifting) makes Mispa i2 different from other conventional nephelometers.
Mispa i2 comes with smart card calibration for all parameters, thus saving time and cost. Also, the rate-based nephelometry technique in Mispa i2 helps in reducing false reporting through the prozone detection feature.
Working principles of Mispa-i2 specific protein analyzer
Mispa i2 works based on the combination of two methodologies - one is rate-based nephelometry, and the other is immunoturbidimetry. Hence the analyzer has 2 detectors namely, the nephelometry detector and a turbidimetry detector.
Rate based nephelometry method offers high sensitivity and the immunoturbidimetry method offers high linearity. Thus, the dual measuring technique offers a wide measuring range and ensures accurate results at all protein concentrations.

Each reagent pack of Mispa i2 comes with a smart card which contains lot-specific calibration data. Mispa i2 offers automated smart card calibration. The smart card calibration feature omits the need for a lengthy calibration process and reduces manual calibration errors. It also saves calibration costs, and reagent costs, and saves time. Hence this feature is quite beneficial for parameters like HbA1c, CRP, RF, etc. which require multipoint calibration.
- Prozone Detection System – Reduces the chance of false reporting
The rate-based nephelometry technique used in Mispa i2 helps in prozone detection and thus reduces false reporting chances. Mispa i2 reads the rate of absorbance continuously, and any repetition of similar absorbance at two different time instants is equivalent to the excess of antigen or antibody in the reaction cuvette (prozone or Kendall effect).

Periodic maintenance of the unit is to be performed by the service engineer of Agappe.
- Mispa i2 Serum Protein Analyzer is for indoor use only.
- The installed site should be free of dust and away from direct sun.
- The site should be free of corrosive gas and flammable gas.
- The bearing or the ground should be free from vibration
- Avoid unplugging the R2 Pipette, continuous plugging and unplugging can harm the sensor pipette socket
- Cover the cuvette holder with an opaque cap when not in use, avoid water spillage.
- Cover the instrument with the provided dust cover when not in use.
